Growth hormone-releasing peptide-6 (GHRP-6), a synthetic 6-amino-acid peptide, is the first in vitro and in vivo active hexapeptide that acts directly on the pituitary gland to specifically stimulate growth hormone (GH) release in vivo and in vitro in a broad range of species . Human Growth Hormone (HGH) in the human body has host of beneficial effects such as decreased body fat, increased muscle, and increased strength and stamina. HGH secreted by the body then causes the liver to secrete the highly anabolic hormone known as IGF-1. IGF-1 also contributes greatly to the body's ability to burn fat and build muscle. Thus, in several studies, when subjects were given GHRP-6, their muscle mass increased and their body fat was reduced significantly. Bodybuilders and athletes have recently started using GHRP-6 in an effort to build more muscle and burn more fat.
Effects
Both Growth Hormone as well as IGF-1 also has several beneficial effects on muscle and bone tissue growth, anti-aging and fat lossing. It would be expected that by using GHRP-6, users would experience the full spectrum of benefits typically seen with the use of HGH and possibly the concurrent use of both HGH and IGF-1. Some data from animal experiments show GHRP-6's ability of GH-releasing stimuli. In 14-d-old rats GHRP-6 (32-600 micrograms/kg, s.c.) induced a marked and dose-dependent rise in plasma GH concentrations, maximal stimulation occurring with the dose of 300 micrograms/kg [7]. HGH secretion was challenged with two secretagogues, postulated to act through different pathways: Growth Hormone Releasing Hormone (GHRH), the hypothalamic physiological GH-stimulus, acting directly at the pituitary, and GHRP-6, a synthetic hexapeptide belonging to the family of GH-Releasing Peptides (GRPs) that binds the Ghrelin receptor in a specific manner[10], and acts both at the hypothalamic and pituitary level [11]. GHRP-6 induces GH releasing in a specific and dose-related manner through mechanisms different from those of GHRH. GHRP-6 is more efficacious than GHRH, and a striking synergistic action on GH release is observed when GHRP-6 and GHRH are administered simultaneously. It has recently been reported that the combined administration of GHRH plus GHRP-6 elicited a powerful GH discharge [12]. Based on such a synergistic action, it has been hypothesized that GHRP-6 acts through a double mechanism by actions exerted both at the pituitary and hypothalamic levels. Further results provide evidence that the GH-stimulatory effects of GHRP-6 are achieved through activation of the PI second messenger system and thus support earlier findings that PKC and Ca2+ play central roles in mediating the effects of GHRP-6 [13].Dosage and Administration
It is recommended to reconstitute the lyophilized GHRP-6 in sterile 18M?-cm H2O not less than 1000mcg/ml, which can then be further diluted to other aqueous solutions. Either by SC or IM, 1000mcg/day will work well to bodybuilders.Differences between GHRP-6 and HGH
GHRP-6 is an extremely potent stimulant of GH secretion in vivo and in vitro. At this point, we can say that GHRP-6 takes effect indirectly and should be considered showing effect not as fast as HGH. But the advantage of GHRP-6 is that GHRP-6 reliably stimulate GH secretion after iv, sc, intranasal, and even oral administration in humans regardless of age or sex.
